
Each tablet of Sizopin contains Clozapine BP 100mg.
Clozapine, a tricyclic dibenzodiazepine is an atypical antipsychotic. It differs from other antipsychotic medications by its profile of binding to dopamine receptors and its effect on various dopamine mediated behaviours. It interferes with the binding of dopamine at D1,D2,D3 and D5 but has a high affinity for the D4 receptors. More preferentially,it is more active at limbic than at striatal dopaminergic receptors.Thus, cloxapine does not produce catalepsy or extrapyramidal side effects.It has an antiadrenergic,anticholinergic, antihistaminic and antiserotonergic action.
Clozapine is well absorbed with an average steady peak plasma concentration of 319ng/ml occurring at an average of 2.5hours after dosage.
Indications:
Severely ill schizophrenic patients who fail to respond adequately to standard psychotic treatment.
Usage/Dosage:
Initial recommended dose: 12.5mg(half tablet of 25mg) once or twice daily. Increment of 25-50mg/day can be given to have a target dose of 300-450mg/day
Daily dosing should not exceed 900mg/day.
Re-initiation of treatment:
Treatment can be re-initiated with 12.5mg once or twice daily.
Peadiatric use:Safety and effectiveness not yet established.
Elderly:Starting dose of 12.5mg on the first day which can be increased to 25mg/day.
Special patients group:Initial dose of 12.5mg on first day with dosage increment made slowly.
